Successful peptide purification depends on selecting the right stationary phase. This application note evaluates the separation of insulin from small-molecule formulation components using flash chromatography and preparative HPLC on the BUCHI Pure Excellence C-950. The study compares different stationary phases and demonstrates how pore size, surface area and stationary-phase chemistry affect chromatographic performance.
Results show that conventional C18 phases did not achieve baseline separation, while wide-pore stationary phases significantly improved the separation of insulin from formulation excipients. The findings highlight the importance of stationary-phase selection when purifying peptides and other biomolecules.
